Protein amyloid aggregate: Structure and function

چکیده
Protein amyloid aggregation has been widely observed to occur and plays important roles in both physiological processes pathological diseases. Remarkably, aggregates assembled by native proteins gain a variety of different biological activities, which cannot be adopted the unassembled protein alone. Thus, it is investigate molecular basis self-assembly how aggregated structure determines its function. In review, we firstly introduce our structural knowledge on undergo conformational transition assemble into aggregate, with main focus fibril, major species aggregate. Then, elaborate structures fibrils enable them fulfill highly diverse functions either or condition. Furthermore, discuss polymorph very unique feature implication understanding structure-function relationship fibrils. Finally, point out importance applying integrating new approaches for deepening study highlight potential designing fibril-based functional bio-nanomaterials application.

The amyloid-beta precursor protein: integrating structure with biological function.
Proteolytic processing of the amyloid-beta precursor protein (APP) generates the Abeta amyloid peptide of Alzheimer's disease. The biological function of APP itself remains, however, unclear. In the current review, we study in detail the different subdomains of APP and try to assign functional significance to particular structures identified in the protein.
متن کاملIslet Amyloid Polypeptide: Structure, Function, and Pathophysiology
The hormone islet amyloid polypeptide (IAPP, or amylin) plays a role in glucose homeostasis but aggregates to form islet amyloid in type-2 diabetes. Islet amyloid formation contributes to β-cell dysfunction and death in the disease and to the failure of islet transplants. Recent work suggests a role for IAPP aggregation in cardiovascular complications of type-2 diabetes and hints at a possible ...
متن کاملAmyloid precursor protein trafficking, processing, and function.
Intracellular trafficking and proteolytic processing of amyloid precursor protein (APP) have been the focus of numerous investigations over the past two decades. APP is the precursor to the amyloid beta-protein (Abeta), the 38-43-amino acid residue peptide that is at the heart of the amyloid cascade hypothesis of Alzheimer disease (AD). Tremendous progress has been made since the initial identi...
متن کاملStructure and Synaptic Function of Metal Binding to the Amyloid Precursor Protein and its Proteolytic Fragments
Alzheimer's disease (AD) is ultimately linked to the amyloid precursor protein (APP). However, current research reveals an important synaptic function of APP and APP-like proteins (APLP1 and 2). In this context various neurotrophic and neuroprotective functions have been reported for the APP proteolytic fragments sAPPα, sAPPβ and the monomeric amyloid-beta peptide (Aβ). APP is a metalloprotein ...
متن کاملSimulations of reversible protein aggregate and crystal structure.
We simulated the structure of reversible protein aggregates as a function of protein surface characteristics, protein-protein interaction energies, and the entropic penalty accompanying the immobilization of protein in a solid phase. These simulations represent an extension of our previous work on kinetically irreversible protein aggregate structure and are based on an explicit accounting of th...
